Insert contains internal PstI and BamHI sites. Restriction digests of the clone give the following sizes (kb): HindIII--4.5, 2.4; PstI--5.2, 1.9; BamHI--5.8, 1.1. Expresses the gene product under the control of the Escherichia coli trp promoter. The vector was constructed by inserting an EcoRI/ClaI fragment with Escherichia coli trp promoter and RBS into pBR322. A HindIII fragment of about 2.2 kb containing the gene with poly(A) signal was cloned into the HindIII site. |
