REFERENCE STRAIN: Collaborative Report on Rhinoviruses, Nature No. 5078, 213:761, 1967. Also for types 13-17: Panel for Picornaviruses, Melnick, J. et al., Science 141:153, 1963. |
Effect on Host |
CPE, rounding and sloughing |
Recommended Host |
WI-38 cells (ATCC® CCL-75™) |
Growth Conditions |
Temperature: 33.5°C
Recommendations for Infection: Plate cells 16-24 hours prior to infection and infect when cultures are 80-90% confluent. Remove medium and inoculate with a small volume of virus (e.g. 1 mL per 25 cm2) diluted to provide an optimal MOI (e.g. 0.1). Adsorb 1-2 hours at 33.5°C in a humidified 5% CO2 atmosphere, rocking continuously. End adsorption by adding virus growth medium.
Incubation: 2-4 days at 33.5°C in a humidified 5% CO2 atmosphere, rocking continuously, until CPE is progressed through 80% of the monolayer. |
